Mechanism of Action of Pioglitazone 15mg:

Pioglitazone Is As A Potent And Highly Selective Agonist For The Peroxisome Proliferator Activated Receptor-Γ (PPARγ). Activation Of These Receptors Promotes The Production Of Gene Products Involved In Lipid And Glucose Metabolism. It Also Improves Insulin Response To Target Cells W/O Increasing The Pancreatic Secretion Of Insulin. Onset: Delayed. Absorption: Rapidly Absorbed. Bioavailability: >80%. Time To Peak Plasma Concentration: Approx 2 Hr. Distribution: Volume Of Distribution: 0.63 L/Kg. Plasma Protein Binding: >99% (Mainly To Albumin). Metabolism: Extensive Hepatic Metabolism Via CYP2C8 And CYP3A4 Isoenzymes To Its Active And Inactive Metabolites. Excretion: Via Urine (15-30%); Faeces (As Metabolites). Elimination Half-Life: 3-7 Hr (Parent Drug).

Dosage

Missed Dose

Take the missed dose as soon as you remember. If it is time for the next scheduled dose then the missed dose can be skipped. Do not take two doses within a day to make up for the previous day's missed dose.

Overdose

Contact your doctor immediately if an overdose is suspected. Appropriate supportive measures should be initiated immediately if the symptoms are severe.

Warnings for special population

Pregnancy - This medicine is not recommended for use by women who are pregnant or are planning a pregnancy in the near future. Use of insulin is most preferred during early and mid-stages of pregnancy.

Breast-feeding - This medicine is not recommended for use in women who are breastfeeding. The risk of adverse effect is very high in infants and hence the potential benefits and risks should be considered before using this medicine.

Type I diabetes mellitus - This medicine is not recommended for use in patients suffering from type 1 diabetes mellitus since it is not effective in treating this condition.

Heart Failure - This medicine should be used with caution in patients having a mild heart failure. Use of this medication is not recommended if the condition is moderate to severe.

Bone fractures - This medicine may increase the risk of fractures in some patients. This risk is especially higher in female patients. Close monitoring of bone health is recommended during treatment with this medicine. Replacement with a suitable alternative may be required based on the clinical condition.

Bladder cancer - Use of this medicine has been associated with an increased risk of bladder cancer. It may also worsen the condition of patients suffering from bladder cancer.

Pediatric use - This medicine is not recommended for use in patients less than 18 years of age.
