The proposed mechanisms of action of Lenalidomidein multiple myeloma (MM) include direct cytotoxicity via induction of apoptosis (A); inhibition of cell adhesion molecules such as intercellular adhesion molecule 1 (ICAM-1) and vascular cell adhesion molecule 1 (VCAM-1), which reduces the growth signaling of MM cells (B); inhibition of growth signals that promote bone marrow angiogenesis, such as vascular endothelial growth factor (VEGF), tumor necrosis factor α (TNF-α), and interleukin (IL)-6 (C); and stimulation of the T-helper cells, which increases production of IL-2 and interferon-γ (IFN-γ) and consequently augments natural killer (NK) cell activation and NK cell-dependent cytotoxicity (D).
