Description

Amantadine hydrochloride is an antiviral that is used in the prophylactic or symptomatic treatment of influenza A. It is also used as an antiparkinsonian agent, to treat extrapyramidal reactions, and for postherpetic neuralgia. The mechanisms of its effects in movement disorders are not well understood but probably reflect an increase in synthesis and release of dopamine, with perhaps some inhibition of dopamine uptake.

Amantadine Hydrochloride is the hydrochloride salt of amantadine, a synthetic tricyclic amine with antiviral, antiparkinsonian, and antihyperalgesic activities. Amantadine appears to exert its antiviral effect against the influenza A virus by interfering with the function of the transmembrane domain of the viral M2 protein, thereby preventing the release of infectious viral nucleic acids into host cells; furthermore, this agent prevents virus assembly during virus replication.

Uses

This medication is used to treat Parkinson's disease. It is also used to treat certain movement disorders caused by some drugs (extrapyramidal reactions). This medication is thought to work by restoring the balance of certain natural substances (neurotransmitters) in the brain.

Different brands of this medication may have different uses. Do not change brands of this medication unless directed by your doctor.

Overdose

If someone has overdosed and has serious symptoms such as passing out or trouble breathing, call 911. Otherwise, call a poison control center right away. US residents can call their local poison control center at 1-800-222-1222. Canada residents can call a provincial poison control center. Symptoms of overdose may include fast/irregular heartbeats, severe drowsiness, shortness of breath, change in the amount of urine, mental/mood changes (e.g., anxiety, aggression, confusion, hallucinations), seizures.

Influenza A Prophylaxis

SYMMETREL (amantadine hydrochloride) is indicated for chemoprophylaxis against signs and symptoms of influenza A virus infection. Because SYMMETREL (amantadine hydrochloride) does not completely prevent the host immune response to influenza A infection, individuals who take this drug may still develop immune responses to natural disease or vaccination and may be protected when later exposed to antigenically related viruses. Following vaccination during an influenza A outbreak, SYMMETREL (amantadine hydrochloride) prophylaxis should be considered for the 2- to 4-week time period required to develop an antibody response.

Amantadine may interfere with the effect of certain vaccines, such as flu vaccine inhaled through the nose. However, you may get a flu shot (flu vaccine given by injection) if recommended by your doctor.
